Sanger sequencing of bacteria and phages

Traditional sequencing of bacteria or phages requires preparation of plasmids. CD Genomics continues to optimize the sequencing process to advance genomic research. Reduce the experimental process and improve experimental efficiency. Our service helps customers to directly sequence bacteria (colonies, glycerol bacteria and fresh bacterial) and phages (or spots) without plasmid preparation, even when customers provide fewer samples (as low copy), it does not affect the experimental results, and our sequencing results are still valid.

Our services can be used in the following studies

  • shRNA cloning and library screening. No need to prepare plasmids, direct sequencing of colonies is possible.
  • Antibody phage display library validation. Direct sequencing of phage for faster screening and confirmation.
  • DNA methylation analysis. Direct sequencing of colonies after PCR product cloning, saving a lot of time and obtaining high quality sequences.

Instructions for customers to send samples

  • Glycobacteria. Transfer the appropriate volume of bacterial solution to 1.5 mL centrifuge tube or 96-well PCR plate (sterile, no nuclease), centrifuge at 14000 X g for 1 min at room temperature, discard the medium, depending on the number of cells, place the bacterial precipitate quickly in liquid nitrogen for 0.5-3 h freezing, then transfer to -80 °C or liquid nitrogen for long-term storage. Note the type of medium, and the samples were shipped on dry ice.
  • Plate colonies. Select the colonies to be sequenced and mark them well. Please seal the plates with sealing film before sending the samples.
  • Phage supernatant. Submit 50 uL of phage supernatant in 8 PCR tubes or 1.5 mL centrifuge tubes, labeled with sample number.
  • Phage spot. Pick the phage spot that is prepared for sequencing and mark it well. Please make sure to seal them with sealing film when sending the samples.
