DNA fingerprinting vs. DNA barcoding

  • DNA fingerprinting, also known as DNA profiling or genetic fingerprinting, is primarily used for plant varieties and strains identification. Our DNA fingerprinting combines SNP genotyping technology for variety specificity, authenticity identification and seed purity identification. We focus on repeating units (e.g., STRs or VNTRs) that exhibit polymorphisms among individuals.
  • DNA barcoding also involves DNA analysis based on PCR and sequencing technologies. While it relies on standardized regions of the mitochondrial or chloroplast genome for biodiversity studies, taxonomy, ecological research, and species identification. Commonly used regions include the mitochondrial cytochrome c oxidase subunit 1 (COI) gene for animals and the rbcL or matK genes for plants.

Samples requirements

Genomic DNA ≥ 15 μL with a concentration between 50-100 ng/μL.
OD260/280 for 1.8-2.2.
Tested for undegraded DNA bands through 2% agarose gel electrophoresis is preferred.
Plant material Transport fresh tissues with leaves silica-dried, maintaining their integrity on dry ice for optimal results.

Service workflow

After sample reception, our meticulous workflow begins with the extraction of genomic DNA from the provided samples. Subsequently, this genomic DNA serves as the template for PCR amplification, utilizing specialized DNA barcode primers. The resulting PCR products undergo a purification process before advancing to the DNA sequencing phase.

Once the sequencing is completed, the spliced sequence files are subjected to a robust comparison using the NCBI Blast program, aligning the sequences with data stored in the extensive NCBI database. This critical step allows us to identify the species with the highest sequence similarity to the target species under investigation, thus delivering a precise and reliable identification result.
