Chromatin immunoprecipitation (ChIP) is used to analyze interactions between genomic DNA and histones, transcription factors, or other chromatin-associated proteins. CD Chromatin Immunoprecipitation (ChIP) Assay Kit provides a 16-reaction reference format covering cell fixation, chromatin preparation and fragmentation, antibody-based immunoprecipitation, washing, elution, cross-link reversal, and DNA purification. Reference reaction conditions use 10-30 µg chromatin per immunoprecipitation. Chromatin input, fixation, fragmentation, and antibody amounts should be optimized according to sample type and target abundance. Purified ChIP DNA can be analyzed by qPCR or used for sequencing library preparation. The standard workflow comprises fixation, chromatin preparation, chromatin fragmentation, immunoprecipitation, washing and elution, cross-link reversal, DNA purification, and downstream qPCR or sequencing analysis.
